This is a working overview of IGF-1, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-10-06. Anything still debated is marked as such rather than presented as settled.
Whether the drug improves hard clinical outcomes is not settled. No completed trial has shown a reduction in heart attacks or strokes among treated patients, although a dedicated cardiovascular outcomes study has been discussed in the literature. Investigators have also examined hepatic fat in people with HIV and fatty liver disease, cognitive measures in small cohorts, and changes in bone density. Regulatory labeling emphasizes monitoring of insulin-like growth factor 1 because supraphysiologic levels raise questions about tissue growth, and the clinical significance of that signal remains an open question rather than a demonstrated harm.
Binding of tesamorelin to the growth hormone-releasing hormone receptor on anterior pituitary somatotrophs activates a Gs protein pathway, raises cyclic AMP, and triggers release of stored growth hormone into the bloodstream. Because the analogue resists dipeptidyl peptidase-4, its plasma residence time exceeds that of native GHRH, producing a larger and more sustained secretory signal. The released growth hormone then acts on the liver and peripheral tissues to raise insulin-like growth factor 1, which feeds back on the hypothalamus and pituitary. This axis explains both the intended effects on fat distribution and the biological markers used to track them.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin binds the growth hormone–releasing hormone receptor on pituitary somatotroph cells. The receptor signals through the Gs protein, raising intracellular cAMP and activating protein kinase A. That cascade triggers release of stored growth hormone in pulses rather than a steady stream. Because the drug acts at the receptor that normally controls this process, its effect depends on the body's own signaling architecture rather than on a synthetic pathway. The resulting hormone profile reflects the timing of each pulse, not only its size.
Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.

Mitoquinone mesylate (MitoQ) is a synthetic analogue of coenzyme Q10 which has antioxidant effects. It was first developed in New Zealand in the late 1990s. It has significantly improved bioavailability and improved mitochondrial penetration compared to coenzyme Q10, and has shown potential in a number of medical indications, being widely sold as a dietary supplement. A 2014 review found insufficient evidence for the use of mitoquinone mesylate in Parkinson's disease and other movement disorders. A 2025 British study found that diabetes patients who took daily mitoquinone, a widely available antioxidant, as well as their standard treatment had healthier hearts after four months than those not given the supplement. Mitoquinone was able to reverse the early signs of diabetes-related heart failure over the course of the study.
== Tissue distribution == RNA expression charts show highest expression in lung and adipose tissue in humans. Cell types that express the highest levels of CALCRL include oligodendrocyte precursor cells, endothelial cells, lymphatic endothelial cells, adipocytes, endometrial stromal cells, as well as dendritic cells.
The reduced sensations can lead to injuries for either partner and their absence can make it impossible to have sexual pleasure through stimulation of the penis. Since the problems are caused by permanent nerve damage, preventive treatment through good control of the diabetes is the primary treatment. Some limited recovery may be possible through improved diabetes control. Erectile dysfunction is the inability to develop and maintain an erection sufficiently firm for satisfactory sexual performance. Diabetes is a leading cause, as is natural aging. A variety of treatments exist, most notably including the phosphodiesterase type 5 inhibitor drugs (such as sildenafil citrate, marketed as Viagra), which work by vasodilation. Priapism, a form of persistent genital arousal disorder, is a painful and potentially harmful medical condition in which the erect penis does not return to its flaccid state. Priapism lasting over four hours is a medical emergency. The causative mechanisms are poorly understood but involve complex neurological and vascular factors. Potential complications include ischaemia, thrombosis, and impotence. In serious cases the condition may result in gangrene, which may result in amputation. However, that is usually only the case if the organ is broke out and injured because of it. The condition has been associated with a variety of drugs including prostaglandin. Contrary to common knowledge, sildenafil (Viagra) will not cause it. Lymphangiosclerosis is a hardened lymph vessel, although it can feel like a hardened, almost calcified or fibrous, vein.

In the late autumn of 1980, having spent several years on spiritual pursuits and then gradually returning to music (including playing guitar for David Bowie, Brian Eno, Peter Gabriel and Daryl Hall; pursuing an experimental solo career; and leading the instrumental new wave band The League of Gentlemen), Fripp decided to form a new "first division" rock group. Having recruited Bruford as drummer, Fripp then asked the American singer and guitarist Adrian Belew, who had previously played with Bowie and Frank Zappa, to join; this was the first time Fripp would actively seek collaboration with another guitarist in a band, and therefore indicative of his desire to create something unlike any of his previous work. After touring with Talking Heads, Belew agreed to join and also become the band's lyricist. Bruford's suggestion of bassist Jeff Berlin of Bruford was rejected as Fripp thought his playing was "too busy", so auditions were held in New York; on the third day, Fripp left after roughly three auditions, only to return several hours later with session bassist Tony Levin (who got the job after playing a single chorus of "Red"). Fripp later confessed that, had he known that Levin (whom Fripp had played with in Peter Gabriel's group) was available and interested, he would have selected him without holding auditions. Fripp named the new quartet Discipline, and they went to England to rehearse and write new material. They made their live debut at Moles Club in Bath, Somerset on 30 April 1981, and completed a short UK tour supported by the Lounge Lizards.

Tea is an aromatic beverage prepared by infusion of hot or boiling fresh water with fresh or cured leaves of Camellia sinensis, an evergreen shrub native to East Asia that originated in the borderlands of southwestern China, northeast India and northern Myanmar. Tea has a stimulant effect in humans primarily due to its caffeine content. After plain drinking water, tea is the most widely consumed beverage in the world. There are many varieties of tea according to differences in cultivars, processing and blends; some have a cooling, slightly bitter and astringent flavor, while others have taste profiles that include sweet, nutty, floral or grassy notes. Traditionally, tea is made by steeping loose tea leaves in hot water (usually in a teapot or teacup) for a period of time, or in the case of compressed and fermented tea, via decoction (simmering) in a kettle. Convenient preparations, such as tea bag, infuser and press as well as powdered instant tea such as matcha, konacha and chai latte, are also common. Tea is most commonly consumed as a hot beverage in a plain hot water infusion, although it can also be in mixed drink forms, such as milk tea and butter tea, sometimes sweetened with added sugar or sugar substitutes, or alternatively flavored with salt and spices. In modern times, tea can also be served as a cold beverage in the form of iced tea or in soft drink mixtures such as bubble tea. Early credible record of tea drinking dates to the 3rd century AD, in a Chinese medical text written by Eastern Han dynasty physician Hua Tuo.
== Research career == After obtaining his Ph.D., his first job was in research for Parke, Davis and Company, and his first task was to isolate the hormone associated with the thyroid gland. He continued this research at St. Luke's Hospital in New York until 1914. He was appointed Head of the Biochemistry Section in the Graduate School of the Mayo Foundation, and the following year he was appointed as the Director of the Division of Biochemistry. Kendall made several significant contributions to biochemistry and medicine. His most important discovery was the isolation of thyroxine, although it was not the work for which he received the most accolades. Along with associates, Kendall was involved with the isolation of glutathione and determining its structure. He also isolated several steroids from the adrenal gland cortex, one of which was initially called Compound E. Working with Mayo Clinic physician Philip Showalter Hench, Compound E was used to treat rheumatoid arthritis. The compound was eventually named cortisone. In 1950, Kendall and Hench, along with Swiss chemist Tadeus Reichstein were awarded the 1950 Nobel Prize in Physiology or Medicine for "their discoveries relating to the hormones of the adrenal cortex, their structure and biological effects." His Nobel lecture focused on the basic research that led to his award, and was titled "The Development of Cortisone As a Therapeutic Agent." As of the 2010 awards, Kendall and Hench were the only Nobel Laureates to be affiliated with Mayo Clinic.

University of Liège professors Joseph Martial and André Renard established Eurogentec in 1985. Prior to establishing Eurogentec, Martial studied genetic engineering at University of California, San Francisco. Martial then returned to Europe and helped commercialize certain genetic engineering research products of the University of Liège. The company began working out of the university's laboratories and focused on developing a genetically-engineered vaccine to enhance fish breeding. Initially, the company focused on the development of a genetic engineering-based vaccine tailored for enhancing fish breeding. In 1987, Eurogentec started to distribute Oligonucleotides for Research laboratories. The company's first years were difficult but in the very early 1990s, Jean-Pierre Delwart, economist from Brussels University, boarded the ship and put Eurogentec in a favorable current.
Germany, the birthplace of decaffeinated coffee, is home to several decaffeination plants, including the world's largest, Coffein Compagnie. Over half of the decaf coffee sold in the US first travels from the tropics to Germany for caffeine removal before making its way to American consumers. Coffee manufacturers recover the caffeine and resell it for use in soft drinks and over-the-counter caffeine tablets. The efficiency and selectivity of caffeine removal depend on the decaffeination method used. Water-based processes and supercritical carbon dioxide extraction are generally more selective, as they remove caffeine while preserving a larger proportion of volatile flavor compounds compared to some organic solvent methods. Extraction of caffeine from coffee, to produce caffeine and decaffeinated coffee, can be performed using various solvents. Following are main methods:

The Samoyed ( SAM-ə-yed or sə-MOY-ed; Russian: самое́дская соба́ка, romanized: samoyédskaya sobáka, or самое́д, samoyéd) is a breed of herding dog with a thick, white, double-layered coat. They are spitz-type dogs which take their name from the Samoyedic peoples. Descending from the Nenets Herding Laika, they are domesticated animals that assist in herding, hunting, protection and sled-pulling.
== Nitrogen-13 == Nitrogen-13 (13N) has a half-life of a little under ten minutes. It is produced in the atmosphere when gamma rays (for example from lightning) knock neutrons out of nitrogen-14. 13N decays to 13C, emitting a positron. The positron quickly annihilates with an electron, producing two gamma rays of about 511 keV. After a lightning bolt, this gamma radiation dies down with a half-life of 10 minutes, but these low-energy gamma rays go on average only about 90 metres through the air, so they may only be detected for a minute or so as the "cloud" of 13N and 15O floats by, carried by the wind. Nitrogen-13 plays a significant role in the CNO cycle, which is the dominant source of energy in main sequence stars more massive than 1.5 times the mass of the Sun. Nitrogen-13 is used in positron emission tomography in the form of 13N-labelled ammonia, for example for myocardial perfusion imaging. It can be produced with a medical cyclotron, using a target of pure water with a trace amount of ethanol. The reactants are oxygen-16 (present as H2O) and a proton, and the products are nitrogen-13 and an alpha particle (helium-4):
=== Liposomes === Liposomes are structures which consist of at least one lipid bilayer surrounding an aqueous core. This hydrophobic/hydrophilic composition is particularly useful for drug delivery as these carriers can accommodate a number of drugs of varying lipophilicity. Disadvantages associated with using liposomes as drug carriers involve poor control over drug release. Drugs which have high membrane-permeability can readily 'leak' from the carrier, while optimization of in vivo stability can cause drug release by diffusion to be a slow and inefficient process. Much of the current research involving liposomes is focused on improving the delivery of anticancer drugs such as doxorubicin and paclitaxel.

It mimics a natural hypothalamic signal that tells the pituitary to release growth hormone. The result is a rise in circulating growth hormone and, indirectly, in insulin-like growth factor 1. Over weeks of treatment this shift is associated with a selective decrease in fat stored inside the abdomen.
The primary measure is usually a cross-sectional abdominal scan that separates internal fat from fat just under the skin. Waist circumference and body weight are recorded as secondary measures because they are easy to obtain but do not distinguish the two fat compartments. Hormone and metabolic blood tests are collected alongside the imaging.
Available follow-up data indicate that visceral fat drifts back toward pretreatment levels once injections stop. The change is therefore best described as treatment-dependent rather than permanent. Investigators continue to debate whether intermittent or repeated courses would preserve any benefit.
It acts on the growth hormone–releasing hormone receptor, a Gs-coupled receptor found on pituitary somatotroph cells. Activation raises cAMP and prompts pulsatile hormone release.
